What is Semax?
Semax is a 7-amino acid peptide derived from a fragment of the complex structure of adrenocorticotropic hormone (ACTH(4-10)).
Semax acts as a modulator of neurotransmitters and neuropeptides in the brain, including dopamine, serotonin, and brain-derived neurotrophic factor (BDNF). The peptide thus plays a key role in transmitting nerve impulses from one neuron to another, to muscle cells or glandular cells, in the growth and survival of neurons, and in cognitive processes. This profile of Semax’s action can help combat mental and physical fatigue, leading to improved cognitive performance and endurance.
Semax exhibits positive effects on brain function due to:
- Neuroprotective action: Semax protects and preserves the structure and function of neurons and can reduce the rate at which neurons are lost over time, thus maintaining mental and physical performance.
- Nootropic action: Semax can improve cognitive abilities, including learning, memory, attention/focus, and creativity.
- Neuroplastic action: Semax can restore the structure and function of neurons to mitigate brain damage and adapt to new situations or changes in the environment.

Research on Semax
a) Semax and Cognitive Function
In a study on the effect of Semax on cognitive functions, it was discovered that a single application of the peptide (at a dose of 50 microg/kg body weight) causes a maximum 1.4-fold increase in BDNF protein levels, which accompanies a 1.6-fold increase in trkB tyrosine phosphorylation levels, and a 3-fold and 2-fold increase in BDNF and exon III and trkB mRNA levels, respectively, in the hippocampus of the tested subject. The study results clearly suggest that Semax improves brain cognitive functions, including memory and concentration, by modulating the expression and activation of the hippocampal BDNF/trkB system.
b) Semax and Anti-Stress Action
Low levels of brain-derived neurotrophic factor (BDNF) can cause mood deterioration. Semax peptide, with its ability to increase BDNF levels, can effectively help alleviate anxiety and trigger mood-improving processes.
During a study on the peptide, dose-dependent stress-limiting effects were observed. The study used an image of the liver of the tested subject after exposure to severe stress. It was noted that it normalized the liver’s protein synthesis function and serum AlAT activity. The peptide’s anticytolytic action increased with the dose against the background of the relative number of multinuclear and multinucleated cells. The reduction in hepatocyte cytolysis against the background of more intense morphological manifestations of protein synthesis processes indicates the activation of repair processes in the liver parenchyma through enhanced constitutional protein synthesis. The changes that occurred in the liver of the tested subject indicate the anti-stress potential of the Semax peptide.

Figure 1. The study highlighted the liver of the tested subject after exposure to severe stress. Images a and b are from observations after direct stress exposure, showing liver parenchyma damaged by stress. Images c and d are from observations after Semax peptide administration, showing reduced stress-induced damage.
The study thus proves the positive effect of the Semax peptide in mitigating the consequences of stressful situations and reducing the body’s stress.
c) Semax and Nerve Protection
Research on the biological impact of the Semax peptide has shown that it has clear nootropic, neuroprotective, and neurotrophic properties. In the tested subjects, after peptide administration, learning and memory formation were stimulated, and these effects were associated with Semax’s ability to modulate neurotrophin expression. In the next part of the study, Semax’s ability to form stable complexes with copper(II) and prevent copper-induced cytotoxicity in nerve cells was also investigated. Clear nootropic, neuroprotective, and neurotrophic effects were observed, making Semax a precursor for the prevention and treatment of central nervous system pathologies.
